Yes, I have a Sega Genesis controler thats wired for the controls in my cabinet. PSX, Genesis, and Nintendo controlers can all be hacked the same way. There is a site showing how to hack Playstation (PSX) controlers in the Examples section.

In most games wouldn't need start and select on the second pad. I think your setup will work.
