I have an ArcadePC and none of the buttons are configurable so I thought I was out of luck until I found this simple trick:

Launch MAME, press tab to pull up the config screen, select Input General, scroll down to UI General, change this from ESC to any combination of keys you want (I used Player 1 + 2 start) by simply pressing the buttons (MAME accepts combinations of buttons!)
