You can use a utility knife and cut several times untill its deep enoough... then put the cut edge over the edge of a table and break it.

My best advice though, is to get them to cut the main shape. Then but a ROTO-ZIP bit. They look like a drill bit, but are sharper on all sides. They melt through plexi like hot butter!
I used a normal multi-speed drill with a drill holding device so that it kept it level as I cut... but you could do without the holder. Just place the plastic over a larger piece of scrap wood. Drill a few holes for screws and then screw it in place. Then drill the rest out with the rotozip bit.

Other things that work good are hole saws, and
circle cutters... though a bit more expensive - and more likly to shatter the plexi.
