Normally they are 8bit, I heard that if you have a proper VBE/VESA3 card then the tweaked modes are 16bit colour, needless to say it was never 16bit colour for me.
All VsyncMAME modes are 16bit, providing you've got one of the supported cards.
Au Natural MAME, no 16bit Arcade monitor support apart from 640x480 ATI mode or Generic 1280x240 mode.
