I think they keep their assignments, but I've been playing with USB for only a week or so. It seems with my USB card "first" controller to be plugged in is Controller 1 and the second is Controller 2.

Theoretically, you can have as many as ports you have. And you can do LOTS with a hub. Specifically: MAME supports up to eight controllers. I think the InputSprocket software supports eight controllers, too.
