Check the message boards at Mameworld.net, particularly the StretchMAME and compile boards. It's pretty straightforward in DOS. I don't know anyone that's done it in MAME32, but the concept is the same.
In StretchMAME32, it will do this automatically for most games if using a 3-axis joystick, but it's not supposed to.
Also, one word of advice, keep a binary with the nag screens on your system also. I like not seeing the intro screens, but when you add a new game or something malfunctions, it is nice to have the screens for sizing.
