I'd put MAME and its directories, DMPI.EXE, the frontend you use, and the the games use most (i.e. if there's a certian game you play about every time) in a ram drive at boot up. I'd make it at least 12MB with the amount of ram you have. When ever you switch between your most used games, it won't have to hit the harddrive, and will load nearly instantly.

I'll post how to create a ram drive later.
