It depends what MAME version you're using. I've been having the same problems while using some frontends. Try right-clicking on mame.exe, choose "Create Shortcut". Now right-click on the shortcut and choose "Properties". Next click the "Misc" tab and you should see "Windows shortcut keys" at the bottom. Uncheck "Alt+Space". This may or may not work if you're using a frontend. Good luck.
