Issue #1, Control Pannel Clutter. Basically you start to have problems if any given control pannel has more htan 2 joysticks if you want to use something else. I have 4 joysticks on my cp so dont even _think_ about adding a spinner or trackball without using a different CP :)
Issue #2, Max Buttons. If you select keys for this game you should only see 4 buttons, but mame supports up to 8 if you select see keys for all games. Buttons for all games is good for assigning coin to button M & start to button start on the sidewinders etc.
#Issue #3, How Many Buttons. Ok. If you just want to play NG you only have to worry about 4 buttons per player + start & coin (trust me you want a coin button). Also no NG game uses more than 2 joysticks at once (although some dont use any at all). For just neogeo emulation you need buttons to control mame & other emus (esc, reset, enter, pause, volume, tab) + 2 joysticks (go with the happs super 8s IMHO but if you wish to play ONLY fighters go with competition). As far as any given game goes I know of a bunch that use 6. For 4 joysticks, I think their are no games with more than 3, but I put 4 on my cp in hopes of CPS2 emulation (AD&D TOD, Aliens Vs Predator). I also put 7 buttons on players 1 & 3. It looks like this (kinda):

The purpose of this is so its good for SF2 etc. Also the staggered 4th button on the first row is good for block in mk 1 & 2, and Run in MK3 & 4 with block assigned to the far right 2 buttons. The neogeo layout uses 4 buttons, so you use the thumb on the lower button and your three fingers no the top to get a kind of arch. overall my CP looks kinda like this:
