I've set up batch files to run certain emulated games with non-standard button layouts.
In the SMS version of Bank Panic, the SMS controller only has 2 buttons, but the arcade version has 3 shoot buttons, so on the SMS Down is one of the shoot buttons.
I have a 4-button gamepad, so I remapped Down to gamepad button #1, and had to remap SMS buttons A and B to gamepad buttons #2 and #3.
This is in a alternate config file for sms emulator MEKA.
The batch file does a rename swap for the config files, then starts Bank Panic on MEKA, then when MEKA quits, swaps back the config files.
This method is ok for when you know before you start the emulator how you want the buttons remapped, but not if you want to make changes in-game.
ex, for your Gauntlet situation, you'd have to know which character you wanted to play before you started MAME.

I believe some keyboard interfaces can support different config files. I don't know if you can change them in-game though.
One thing that seemed nice about the PowerRamp was it's macro programmability. It'd be nice if you could have that in other keyboard interfaces.
