In theory, yes, a ghosted keystroke would show up when using Keyhook. It just displays the output of whatever signal that it's getting, so if you never got a ghost in Keyhook, you shouldn't get any ghosting in MAME either (of course, provided that in MAME you don't hit any key combinations that you didn't hit while testing it with Keyhook). You sound good to go!
